Description enPsoriatic arthritis is a chronic inflammatory skeletal disease associated with the skin disease psoriasis. PsA is a common disease, but many aspects of this disorder remain enigmatic. Increasing evidence suggests that the primary disease manifestations occur in the attachment sites of ligaments and tendons to the bone. This anatomical zone is called the enthesis. Onset and further development of the disease takes place in the synovio-entheseal complex referring to the tissues in close proximity and in a functional relationship with the actual enthesis. In this project we test the hypothesis that biomechanical stress can activate inflammation in the synovio-entheseal complex. Distant inflammation in the skin or the gut may be a co-triggering factor. Moreover we earlier defined a set of genes associated with mechanical stress in cells derived from the synovioentheseal complex. We will test if overexpression of these genes can also trigger inflammation. To test these concepts we plan a series of in vivo mouse model experiments as well as in vitro cell culture set-ups. The project uses state of the art technologies including lentivirus-mediated gene transfer. Overall, we aim to get new insights into the onset of psoriatic arthritis and may therefore identify novel therapeutic targets.
