Zeb2 staat bekend als een regulator van de expressie van andere genen. Onze hypothese is daarom dat Zeb2 de LSEC-specialisatie bepaalt door regulatie van (LSECspecifieke) genexpressie.

| Description en | Endothelial cells (ECs) are highly heterogeneous across the vascular tree having different properties that are largely transcriptionally regulated. ECs lining liver sinusoids (LSECs) are specialised in transport from and to hepatocytes. Hereto, LSECs are fenestrated and have high endocytic activity. Furthermore, LSECs are ‘zonated’, i.e., they have different characteristics in areas around the portal veins versus areas around the central veins. LSECs are constantly exposed to damaging substances, including drugs and toxins and therefore play a key role in liver disease. To learn more about the transcriptional regulation of LSEC specialisation, I aim to: (i) transcriptionally profile LSECs at different developmental stages until adulthood to obtain stepwise information on TFs involved in LSEC differentiation; (ii) determine the developmental time frame of LSEC zonation and its transcriptional profile and regulation thereof; (iii) transcriptionally profile LSECs at different stages during non-alcoholic fatty liver disease; and (iv) to study the role and underlying mechanisms of Zeb2 as a transcriptional regulator of the LSEC signature in healthy and diseased livers. Unravelling the transcriptional regulation of specific LSEC signatures in physiological and pathological circumstances may deliver invaluable information to generate LSECs for inclusion in bioreactors and to develop strategies for treating liver disease.
